The actual oldest building in Denver is Four Mile House, built in 1859. It was originally four miles away from the main downtown stage stop, hence the name. Now it's in east Denver, roughly at Forest and Leetsdale.

Hard to say for absolute certainty, but the little town of San Luis has the oldest spanish church or mission in Colorado and is considered the oldest town. Pueblo was also one of the earliest settlements. The town of San Luis lies just north of the New Mexico border in the southern portion of the San Luis valley, a land feature so large it can be in fatc seen from the moon! The town lies south of Alamosa. I've been to the church, it's small and dates from the 1500's.
