The majority of the passengers on the Mayflower were English religious Puritans who had been living in The Netherlands in order to enjoy the religious freedom that was a available to them there but not in England. However, they did not like the cultural assimilation that was impacting their community, so they obtained from the King of England a grant of lands to settle in America. They hired the Mayflower to take them there. The voyage occurred because that was how they traveled to America.

Pilgrims! They were from the U.K., and they came over here on the mayflower. They came because they were sick of not having any freedom in the U.K. They didn't even get to choose their religion, King George chose everybody's religion, which was always what religion he believed in. So, a group of them came over on some ships (the main was the Mayflower) to gain new freedom. Then they ran into the united states...
Not 100% sure but I believe not. They pretty much accepted anyone that was of their religion.

No there wasn't any ship named the Mayflower in any of Columbus's voyages in 1492. The pilgrims were the people who sailed on the Mayflower to leave England in 1620 from persecution because of their religion.
