The ghosts in the Mario games are called Boo Ghosts (simply referred to as Boo or Boos). When you enter the castle, on either side of the staircase you should see 2 wooden doors. When you go through them and into the hallway, you'll see a big Boo, which runs away if you attempt to chase it. Go through the door it disappears into, and you will end up outside in the courtyard. There are many Boos here. Find the big Boo, and face away from it (this makes it big and able to be killed; if you face Boos they will turn shy, shrink, and are unable to be killed), and when it sneaks up on you, press A (jump) and then R to perform a ground pound to kill it. It will leave behind a birdcage-like object. You can walk into it to enter Course 5: Big Boo's Haunt.
