If you mean the story line of the game, no you can't use Wesker. But you can play as him in Mercenaries, to unlock him you need at least 4 star ranking in the Water Island stage.
You can only play Wesker in the mercenaries. It's pretty sad. It would be cool to play the game from his perspective though.
