Well, there are two ways to go about doing this.
1.) Right off the bat, you can kill the boat captain and steal the key, but I don't recommend this, as it messes up the story arc.
2.) Complete the main storyline in Point Lookout. When you finish and head back to the boat, the girl will be waiting, telling you that the captain is in the engine room. You then enter the room and kill him. But be prepared. It's kind of freaky in there...

You should receive a radio message informing you that the quest is active, all you have to do now is get on the ferry to Point Lookout. You can find the boat south of the Citadel, at the very bottom of the map.

After purchasing andinstalling point lookout from windows (either that be games for windows or xbox live) launch FO3 and play as normal for up to several minutes the player will receive a message upon the HUD describing the Duchess gambit landing in the estuary of the Potomac what ever your level, you can proceed to the boat as normal
A few ways 1. Use the riverboat tickets. 2. Complete The Local Flavor (possibly) or up to the part where you meet that missing girl. 3. Complete all Main quests of Point lookout. Note: i played Point lookout like twice then stopped playing for almost 3 months since i got tons of new games.
