The filing cabinet is behind the teacher's desk in the Detention Room. You need gum and a stink bomb. Collect all 8 comic pieces and give them to the comics shop guy (who will give you the frame for them). Assemble the comics and he will give you some gum.
Use the combination on the comic to open Nate's locker for the "hidden room blueprint". In the Science Lab, use the chemistry set to create a stink bomb (mixing and heating the yellow and blue chemicals).
Chew the gum in the hall and get detention. Use the stink bomb there to run the teacher off. The secret room is behind the filing cabinet, as shown on the blueprint in Nate's locker.
(Note : if you don't have the blueprint, you cannot access the filing cabinet. Go outside and get it, then simply re-enter the detention room - stink bomb is still there).
