Once you activate one of the Totems of Hircine in the Underforge, you should gain a power that can be activated the same way you would use a shout. Make sure you have it selected in your magic menu and then push your powers button.
Also note that Totem of the Hunt gives Detect Life. And if there is nothing around it may not look like anything is happening. Also, Totem of Fear causes the fear effect on enemies, but up to a certain level. If there are no enemies around or their level is too high, it won't do anything.
You can test if things are working by using Totem of Brotherhood to summon two wolf spirits as this one should always work.

You have to go to the Companions in Whiterun. After a few quests they will turn you into a werewolf.

Dawnguard is the expansion that adds the werewolf perk tree, without that, you can't get werewolf perks.
