Okami means god ('kami' meaning god, and 'O' being a respectful prefix)
Ookami means wolf.
There's a slight pronounciation difference with "ookami" having a longer 'o' sound. I believe I read somewhere that the pronouciation is wolf, but the Kanji (on the Japanese cover) is god, hence adding to the pun.
This of course all makes sense since basically the story follows a wolf (Ookami) who is actually a god (Okami).
