Emmet, the weapons dealer's backyard can be found in Willowfield, Los Santos. Just across the road from there is an Ammunation Store.

There are also Ammunation stores situated in these locations:

On the outskirts of Las Venturas in Bone County.

El Quebrados in Tierra Robade.

Fort Carson in Bone County.

Blueberry in Red County.

Palomino Creek in Red County.

Market in Los Santos.

Willowfield in Los Santos

Ocean Flats in San Fierro.

Come-a-Lot in Las Venturas.

The Old Venturas Strip in Las Venturas.

In the mission "Doberman" which takes place fairly early on in the game in Los Santos, CJ gets to enter the first of Ammu-nation gun stores, which is in Downtown, Los Santos. Sweet calls him to deal with a pusher problem in Glen Park, so CJ is required to go to Ammu-nation first to get tooled up, and is directed to go there. The gun store is then unlocked.
