When Ralph blows the conch, it signifies that there will be a meeting and that all should gather round. When a particular person is holding the conch, it means that they are allowed to voice their thoughts an opinions. The conch itself signifies civility and order.

In The Lord of the Flies, during the first assembly Ralph lays down the rule that only the person holding the conch can talk. The conch symbolizes law, order and civilization.
In chapter 5 of "Lord of the Flies," Ralph should have ended his speech when Jack attempted to take the conch, because his failure to do so resulted in a lack of order. At this point in the novel, Ralph has lost much of his authority amongst the group. When he berates the boys during his speech they are disinterested and focus entirely on the the possibility of a beast on the island.
The sound of the conch represents a time to assemble at the platform. That is how they all found each other when Ralph first found the conch, all the boys started coming out of the jungle to see what it was.
