If by harmful you mean dangerous, then it is because they have a natural instinct to hunt down/kill their prey. Like a cat and a mouse - the cat automatically thinks it is food because they recognise the mouse as his food.

A tiger normally won't harm humans. However, because we settle in their habitat, we place ourself in the fridge of a hungry tiger.

They can be harmful to humans. If they smack a human with their paw, even if they're just playing, it could kill them.
