Usually you shouldn't need to express your dogs anal glands manually as the dog will generally do this on their own. Anal glands are expressed naturally when your dog has a bowel movement. If your dog has been scooting on the floor, biting their anus or if there is a rash or swelling of the anus then take your dog to the vet. A infection will not be erradicated by expressing glands manually...this only makes it more comfortable for the dog and does not treat the infection. Medication will be required.
