For security reasons, the President and Vice President usually don't travel together (so if there's an assassination attempt on the President, the VP can be kept safe, in case the president dies and the VP has to take over). That, by itself, would put a lot of strain on a marital relationship, on top of the fact that, under normal circumstances, the president and his or her spouse are usually so busy they hardly ever get to see each other.
The marital status of the President and the Vice-President are not legislated or discussed in the Constitution, so there is nothing to prevent it.
