I suppose it is very possible to cause an unintended problem with the transmission while replacing the rear seal as it involves either removing the transmission, or at least pulling it back. If its an automatic transmission, you would have to remove the torque converter and flex-plate. If something doesnt get properly torqued to specs, then Yes you could cause a huge problem. You also run the risk of damaging the drive-line, slip yoke, Universal joints, etc.... Not the best job for a first timer to do in my opinion.

No, changing the size of your rims and tires will not mess up your transmission. It will mess with your gas mileage and your speedometer though.
From what I heard and from my past experience. Sometimes it does hurt the transmission. What I mean is it depends on the transmission wear and tear. For example; I had a 1990 Nissan Pathfinder SE, Transmission started slipping a little, so I figure it would help. Well to make the long story short. I change transmission filter and fluid and I got 1 more year out of it.

Maybe use some kind of siphon pump... It would be a complete mess trying to remove the transmission oil pan. Or maybe just go in and get a fluid change.

Changing transmission fluid is simple, but a real mess - drop the transmission pan and collect the juice. Change the filter (if there is one) and reattach the pan using a new gasket and refill the fluid. I suggest you take it to a transmission shop to have this done, as they will inspect the transmission and adjust the bands, and keep your driveway and yard from having a nice red stain. I also have heard many tales about problems and leaks immediately after changing the fluid, so I wouldn't change it unless you are well over 100K miles or drive up mountains all the time.
