I Recently had a problem with my 2000 Odyssey sliding door. Initially, the passenger side sliding door would not latch completely. But I did not fix it for sometime -- I would just push it to latch. However, after several months of doing that, the sliding door warning lights on the dashboard came on and I could not open/close the door neither from the inside, outside or via remote.
Posts elsewhere indicated that this worked on both 2000 and 2001 Honda Odyssey vans.
Suggestion #1
1.) Disable the power door switch on the left dashboard.
2.) Remove the power door fuses located under the front passenger leg area.
3.) Manually open and close both sliding doors.
4.) Restore the power door fuses located under the front passenger leg area.
5.) Enable the power door switch on the left dashboard.
6.) Try opening the sliding doors.
Suggestion #2
Remove fuse #13 (clock) for 30 seconds or more from the passenger side fusebox. This will reset the door and the clock.
From Purchasepete - Suggestion #2 - has worked for me, on 2 different 2001 Odysseys !
Suggestion #2 Works fine (and is so SIMPLE!) for 2000 Odysseys too.
The no.1 solution works for my Odyssey 2000, except for me the number of the fuse was 12 instead of 13... Maybe because i have the Canadian version of the van.

A VERY simple alternative to trying all these steps is to simply disconnect your battery! No need to play around with the fuses, just open the hood, unplug the black wire from the battery, wait for a few minutes, put it back on and voila! it works!
It did for me anyways!

Regular maintenance and checks can help prevent such issues.
Try the answers for 2000 sliding power doors. I know they re-made the vehicle in 2000 but it might be relevant still.
To fix a sliding door on a 2005 Honda Odyssey, first, check the door track for any debris or obstructions that may prevent smooth operation. Inspect the door’s rollers and hinges for wear or damage; lubricate them if necessary. If the door is misaligned, adjust the alignment by loosening the screws on the hinges and repositioning the door. If issues persist, consider replacing the door latch or the motor for the power sliding feature.
The automatic sliding door
A disconnected door handle linkage can cause your sliding doors not to open. The locking mechanism may not be fully disengaging, of which will cause your door not to open.
