It would be best to indicate in conversation that you have other gay friends and that you do not have a problem with gay people. Perhaps open that door with a political conversation i.e.: California's recent Prop.8 controversy. If you have a problem with people being gay then you really don't need to know if someone is gay.

Most of the time it is almost impossible to tell what someones sexual preference is unless they tell you, or make it obvious. If you are asking how you tell someone that is straight that you like them in a sexual way, it probably will not go well. You can always defuse the issue from the start by telling them that they don't need to worry about it, you happen to be gay, but you are not trying to get in their pants. If you show them that you are someone they might want to be friends with, being gay will not be that big of a deal. If they can't deal with you being gay, you don't want them as friends anyway.
