Sperm donors are required to collect their semen by masturbating. They are generally provided a private room to collect their donation and sometimes provided pornographic material.

He is given a sterile collection container and instructed to wash his hands, and sometimes his penis, in order to collect a clean semen specimen. Lubrication may be provided. Donors are not to use saliva or any lubricant brought from home, as this may contaminate the semen with bacteria. Any contamination introduced may cause an infection in the female patient ("Recipient") who will use it later. It is not permitted to have a girlfriend/partner "help" the Donor collect the semen specimen.

In the U.S. and Canada, donors are required to collect their donations on the premises of the sperm bank, in order to make sure the semen being stored in the Donor's name is actually his.
