Simply put, a synthesizer is a computer which calculates mathematical functions and plays the result of those functions on an amplifier. It does not use pre-recorded samples, but creates sounds literally from nothing but electricity.

Analog synthesizers contain voltage-controlled oscillators. An oscillator is a circuit that uses positive feedback to create a periodic waveform. A voltage-controlled oscillator varies its frequency according to the (DC) control voltage that it is given. In an analog synth, the keyboard produces a different control voltage for each key that is pressed, so that the oscillator generates the frequency that corresponds to that musical note. The output of the oscillator(s) goes through filters and then to an amplifier, to drive speakers.

Digital synthesizers contain microprocessors, which are programmed to generate a stream of numbers, usually pulled from tables in memory, representing a waveform for a particular pitch. The stream of numbers goes through various algorithms, including filter functions, before being sent to a digital-to-analog converter. The output of the D-to-A converter is an electrical signal that can be sent to an amplifier, to drive speakers.

Software synthesizers are programs that make a general-purpose computer work like a digital synthesizer, as described above. The computer's processor does all the waveform generation and filtering. The stream of numbers is routed to the operating system's audio subsystem, and eventually goes to a D-to-A converter on the computer's sound card, which usually drives powered speakers.
