Restriction enzymes (RE) are endonucleases that isolated mostly from bacteria. It is used to digest or cut the DNA fragment at a very specific locus. REs are highly specific to its substrate for example EcoRI enzyme cut exactly between G and A in the sequence of GAATTC in both strands of DNA. So they are widely used in gene cloning.
