From 1910 until the second world war, it was one Korea but under Japanese rule. From 1897 until 1910 there was the Korean Empire. Before 1897 the Korean Peninsula was united, but it was constantly at war with China, with Japan, etc so a lot of influence came from both countries.
It was only a convenient line to use which was almost halfway down the Korean peninsula. It was used after World War 2 to separate the Russian held north from the area designated to the other allies.
