In tenpin bowling, bowlers who are having difficulty with release, usually related to the ball sticking on the thumb, will sometimes have a very small hole drilled just below the thumb hole, at angle that enters into the lower part of the thumb hole. This is to allow air into the lower region of the thumb hole and prevent a vacuum from taking hold.
If you are trying to make the thumb hole then you just sew a basic button hole (but larger of course)

remove the buttplate screws, and the buttplate. In the large round hole in the stock is the stock bolt. That is unscrewed to remove the stock from the rifle.
I believe that wild dog stocks of Australia are now manufacturing their thumb-hole stock for the .22lr CZ varminter for South Yorkshire Shooting Supplies (rimfiremagic). Wild dog stocks are very nice - might be worth contacting them: they have distributors in the USA and Europe AND do a variety of cool custom paint jobs.

You need to be a bit more specific about WHICH 22 rifle, but there were several that loaded through the buttstock. On those, the hole is where ammo is loaded into the rifle. Hole is typically an elongated teardrop shape.
