from a true gamers perspective, it is my humble opinion that one drive is best. like a chain, a computer is only as fast as its slowest part. and rarely do I feel that the hardrive access time is the "bottleneck". if you have a slow video card then you will see it in the games...does having two harddrives make games run faster? possibly yes. but the difference is not huge. load times will tighten I suppose and if your dead set on extreme gaming performance then you may want a "RAID array hardware setup. which for the most part is TWO harddrives working together. it is faster, more advanced, more complicated and more expensive than just one hardrive. however if your just looking for a solid machine for games. money is better spent on a midrange hardrive and high quality videocard or processor perhaps.
