No.
Hotglue isn't stretchy, and bike tubes, and patches are. Sticking something stretchy onto something stretchy with a stiff glue isn't going to work.
If it is tubeless then patch the tire on the inside. If it has a tube patch the tube.
If the bike is set up properly, with a real fixed hub, then you should be able to do it. Keep in mind that the contact patch on a road bike is quite small, and that you're likely to wear your tire out quickly.
If you have a mountain bike, you can often remove the tube, patch it with a patch kit (auto parts stores and bike shops have them) and put it back in. Those patches don't hold up very well on a road bike with 700C wheels because the air pressure is so high, so you need to replace the tube on those.
In order to find the proper replacement for your bike tire you need to measure the wheel and determine which type of tire your bike uses. You can also take it to a bike repair shop and they can help you pick out a replacement tire.
You can find a bike tire by visiting a local bike shop or even a retail store like Walmart.

use a tire patch kit
yes but you have to have a kit or at least a tire patch
depends on what kind of tire you are talking about like a bike tire or car tire or a motorcycletire slash /dirt bike tire. be more specific next time. bub!
The fork of the bike is where the front tire is placed.
yes it can be patched
It takes very little skill or effort to change a bike tire. Anyone who rides a bike should be able to do it.
