No you cant, if you really wanted to convert it you'd have to change the whole transmission.
Yes you can but, this would be a very expensive job (labor and parts). But if you really want to do this, first locate and purchase a vehicle similar to yours that is automatic and use the vehicle for a donor. Swap all parts over from the automatic donor car into yours.
It also depends on the vehicle. Current vehicles typically will require the ECM to be reprogrammed on top of everything else which needs to be done. A lot of older, all mechanical vehicles, it's not such a task... an SM465 manual swaps out for a 700R4 automatic quite easily. On a more modern vehicle, it's not so easy.

This is a major mechanical project. Remove the clutch and manual transmission and the radiator. Install automatic transmission, new driveshaft, new radiator. You have just spent as much as a car is worth.
