It is used when a person does something even when he has been forewarned about something or, when a person repeats a mistake though he was punished for the same, before.

For example, "In spite of telling him not to come, John came today."

Or, "Mindy went out to play in spite of getting scolded the previous time".

"In Spite Of" also means the same as "despite".
