You take penicillin exactly as your doctor has told you to take it - do not take it if you have not seen a doctor and actually been tested to make sure you have an infection. It's not going to help if you have a virus, and will just make you resistant to it so it won't work in the future. Penicillin is usually taken every 6-12 hours however.

Yes. Penicillin can be taken every four hours. But is is usually not needed. Your physician will give you usually three to four times a day. Even two times a day dose is enough to cure you of strep throat. But some times the patient may forget to take the dose. Then it becomes once a day dose. It is not enough to cure strep throat. So the wise physician will give a dose of penicillin three times a day. So even if one dose is missed, there is no problem.

Yes. You can take amoxicillin every four hours. But then it has most probably no advantage, in most of the infections over taking every eight hours.
