Yes, it can and in most cases it is a few degrees off. Depending on the type of thermostat (old style Mercury-bulb,

bi-metal, or digital) some are very inaccurate and it'll cause the system to run much longer than it should, and turn on/off more frequently. Also, if the thermostat is near a kitchen, direct source of sunlight, or if it is in an area where there is no return-air grille, it will be affected substantially.
