Nope, they don't have to at all. The only place they have to go (but only if they are able to, like financially or healthwise etc.) at least once in their lifetime is Mecca on Hajj trip since Hajj is the 5th and last pillar of Islam.
Yet, people are encouraged to visit the three places: Makkah, Medina and Jerusalem due to their value in Islam and the multiplication of virtues by doing so.
One Raka'a "prayer" in Makkah = 100,000 prayers elsewhere.
One Raka'a "prayer" in Medina = 1000 prayers elsewhere.
One Raka'a "prayer" in Jerusalem = 500 prayers elsewhere.
