If your reloading after the chamber is empty and there are no more cartridges in the magazine, Yes you have to pull back the charging handle all the way and let it go. If your doing a tactical reload and just topping off your mag; No.

If it is one of the recent Colt .22LR AR15 rifles, then the answer is no - they are completely different internally.
If you are unsure how to safely operate your firearm, it would be advisable to get some advice/training from a knowledgeable individual. The salesmen at the gun store can help you understand how to properly use and load your firearm. Put the magazine into the rifle brass side up. Give the magazine a little tug to make sure it is properly seated in the magazine well. To chamber a round, pull the charging handle back (which will pull the bolt back) and let the bolt cycle forward. Hit the forward assist to make sure the bolt is completely forward; otherwise the firing pin will not hit the primer.
