No. When you travel to Puerto Rico, it's like going anywhere within the U.S. All you need is a driver's license or other valid form of photo ID. Puerto Rico is considered a Commonwealth of the United States.

As a Columbian citizen with US resident status, you do not need a passport to travel from elsewhere in the US to Puerto Rico, but you will need proof of your resident status and a government-issued photo ID.

If you are traveling to Puerto Rico from Colombia, or any other location outside the US, you will need your passport to enter the country, and you may what to have proof of your resident status with you as well.
