No. Even with the new Western Hemisphere initiative effective June 1, 2009, travel directly from the US to a US territory does not require a passport.

We are traveling to St. Thomas in March 2008. I called the National Passport Information Center to ask if we needed passports. The representative said you do NOT need a passport to travel any US territories including St. Thomas. NPIC 877-487-2778.

No, a passport is not required because it is considered part of the u.s. territory, so basically you have not left the united states. Although it would be wise to have a passport anyway incase your flight gets diverted, or you decide to go on a boat off the island that could end up outside u.s. territory. No you do not but there are many misinformed Law Enforcement Officials who will attempt to persue you. Ignore them and run if necessary. If it comes down to a court case you will win even without a lawyer.

I just check this out with my travel agent because I am leaving on 6-10-09 by air. She said you do not need a passport because St. Thomas is a U.S. territory.
