You have two options. One is to use a suction device to remove the oil through the fill hole. The other is to remove the diff cover to drain, then reseal and fill.
there is usually a bolt on the inspection plate of the diff and you fill up to the hole where the bolt is.
there is no actual drain plug on the rear diff. the fill plug is on the right side of the diff just above the gusset it takes a 3\8 ratchet to remove it then u need a vacuum pump to remove the 80\90 gear lube otherwise you must remove the rear diff cover to fully drain the rear diff also you will need a new gasket for that as well
Usually there is a fill only & have to remove the cover. When you reinstall just fill to bottom of hole. Plug on cover or top of "pumkin".You need a diff cover gasket & gear oil of course. It's tricky getting a good seal so use a torque wrench & criss cross when the time comes.
front or rear? the front is slightly higher and rear of the drain... the back has a cover you remove to drain the fluid and a fill on the front of the diff..

put a drain pan under the diff, unbolt the diff cover and pry it loose, oil will come out. clean out the debris from the cover, put it back, fill diff thru fill hole on cover until oil starts to come out of the hole. put cover bolt back in place. be sure to torques bolts to spec and to tighten the diff bolts in the proper order to avoid warping the cover.
raise the rear end 8 inches remove fill plug then drain plug, replace copper washer on plugs insert drain plug to 30 ft-lbs, fill by using small siphon hand pump, or angle the bottle, insert fill plug 30 ft-lbs the rear diff holds 1.7 quarts.
The standard capacity is about 2 quarts and you will need to put in a bottle of friction modifier. When you fill it up, it should reach to the bottom of the fill hole, with the vehicle at normal ride height.

You will need to remove the plate on the housing to drain it. There's a fill plug on the front side where the drive shaft hooks to it.
