So, how is it done? It isn't.

benzodiazepines
cocaine metabolite
opiates

methadone
methaqualone
propoxyphene
These types of tests are called immunoassay tests or gas chromotography with mass spectrometry (GC/MS), and will usually use creatinine levels and oxidant screening of chromium and nitrates levels for adulteration detection.
