They show you keys that you can use to activate those menus and items on menus. Pressing and holding the Alt key and then the F key will open the file menu. Pressing X then, would select the Exit option and close Excel. Alt activates the menu bar. You can use these keys to do things quickly. Alt - E - C is Alt, Edit menu, Copy option. So the underlined letter is the letter you type to select that particular option.
The two of them alone don't do anything. Alt will activate the menu bar, which you could then open a particular menu with by hitting the underlined letter of the menu in question. If you hit Alt and Shift together, the only thing that will happen is the menubar being activated, and that is purely because of the Alt. The Shift has no effect.

The F10 key in Excel activates the menus. It does the same as pressing the Alt key by itself. In older versions of Excel, up to Excel 2003, menus were used and the Alt or F10 key activated the menu bar. They both work in versions of Excel from 2007 onwards to activate key tips, so that you can use keyboard combinations for certain activities that would have been on the menus on older versions. You can also use F10 with other keys. SHIFT+F10 displays the shortcut menu for a selected item. ALT+SHIFT+F10 displays the menu or message for an Error Checking button. CTRL+F10 maximizes or restores the selected workbook window.
