Maybe. It depends on the mail program they use and the options they have chosen. On several popular mail programs, the user can choose whether to automatically send return receipts or ask them each time. If they've chosen to be asked, then yeah, they're gonna know. If not, they might or might not notice.

The safe bet is to assume that they WILL know; that way you don't get unpleasantly surprised.
