Two main reasons a car wont start are gas and fire. Is it getting gas to the tbi (carb). Are the plugs getting fire(spark). Track these two down and you will find your problem.

you can as long as you use your TBI heads if you want to use the vortec heads with TBI you will need a special intake manifold or a carb manifold with a adapter to mount your throttle body to the carb manifold heres some more info TBI heads will mount to the vortec block TBI intake manifold has 12 bolts vortec intake manifold has 8 bolts

It is not a bib deal but the problem is that ther is not enough gasoline going through the tbi and it's hoses. I think the hoses or the actual tbi is cloged with some sort of debri. You should take out the tbi and its hoses and clean both of them very good. Hopefully the garbage in the tbi and/or the hoses would have came out!
