Black leopards are a helpful mutation. It allows leopards that live in dense cover to camouflage to the shadowy forest.
This is not entirely accurate, since a spotted leopard hunts just as well, if not better than a melanistic (read: black) leopard. This does not make it anymore helpful than blue eyes as opposed to green. Though undoubtedly beautiful.
Also I should add that, while some do, most leopards live on the savanna and not in forests, and they rely more on the difference in the color of their fur to better camouflage themselves. Nice try though.

The single mutation is in the B subunit of hemoglobin. One point mutation changed one amino acid and in malaria country the resulting trait, sickle cell trait, is very helpful to have in a heterozygous condition.

Not all mutations are harmful. A mutation the give the organism antibiotic resistance, for example, is quite helpful. A different mutation that causes a necessary protein to misfold may result in death. In general mutations that affect proteins that are necessary for life will result in the death of the organism. One such mutation is in the protein p53 which is necessary to prevent a cell from growing uncontrollable (cancer). A mutation in p53 could result in a cell with damaged DNA to reproduce - this is what we call cancer.

Mutation is caused when chunks of chromosomes cut taken or fall off. So the chromosome with the DNA causes a mutation which sometimes is helpful but mosty hard to live with.
