There are many different poisons, which work in many different ways. You would need to name the poison and try to find out exactly how it works. Poisons have one thing in common though; they all interrupt some vital metabolic pathway, hence causing death. For example, for a person to live, he/she needs oxygen. The oxygen is carried from the lungs to the cells by a molecule called Hemoglobin. If carbon monoxide (a respiratory poison) is present in the air, the Hemoglobin in the blood does not bond with the oxygen but bonds instead with the carbon monoxide (the compound formed is more stable). Therefore, respiration slows down, and the individual dies.

That depends on a poison, e.g a neurotoxin shuts down the nervous system, which in turn shuts down the vital organs (...heart, lungs, brain...), and hemotoxins kill blood cells.
