In Brazil, the preferred "you" is "você", in Portugal it is "tu". (some States of Brazil uses "tu")

Como vai você? (how are you doing?) - use of VOCÊ

Como estás? (how are you doing?) - use of TU

Como você está hoje? (how are you today?) - this would be used specially if the person was not feeling well before... to 'check' on their well being.

Como estás hoje? (how are you today?)
