You can't send a private message to someone who isn't following you, but you can send a reply using a at (@) followed by the person's username and your message. You can do this by Mentioning them or by replying to one of their tweets.

Who gets the tweets when you send?
Your tweets will appear on the time line of those people that are following you. Also, anyone who clicks onto your profile will be able to see your tweets if your account is not "protected".
I am following someone on Twitter who's tweets are set to private. My tweets are also set to private. They are not following me but I am following them If I send them replies will they get it?
Yes they will get your tweets. It will appear in their @replies tab.

If your account is private then only people that are following you can see your tweets. It doesn't matter who you are following, they must be following you to view your tweets. So, if you are following someone but they aren't following you then they won't be able to view your tweets.

The only way you would receive these tweets is if they are retweets or if you are following them.
