The most reliable way of sexing a Corn Snake is by 'probing', a metal probe is inserted into the opening of the pouch behind the vent, in females the probe will extend the length of only 1 or 2 subcaudal scales, while in males it will extend 6-8. PLEASE NOTE: THIS PROCEDURE SHOULD ONLY BE CARRIED OUT BY SOMEONE QUALIFIED TO DO SO AND CAN SERIOUSLY HARM AND CAUSE GREAT PAIN TO YOUR SNAKE IF DONE INCORRECTLY!
There's generally two (2) ways you can do so:
1. For most (and i do mean most) snake you can generally "tell" their sex by the length of their tail, from tip to their cloaca (there reproductive hole or sometimes referred to as the "base" of the tail). If the tail is fairly long from tip to the base it would possibly be male, short and stubby from tip to base would possibly be female.
2. On some snakes you can GENTLY place your thumb right underneath the base of the tail (right under the cloaca) and then your index finger right behind for support, as if you were holding a coin in between your thumb and index. Then with your thumb, you bend the tail at the base and then GENTLY (i stress this part) push your thumb towards the upper part of the snake and their should two (2) small things that pop out, if there are, this would be the snakes hemipenis....meaning it's a male.

Snakes are very hard to tell gender wise. If you don't know what your doing it's best to take them to a vet. If you do know what your doing then there are 2 ways, probing or popping. Probing is where you use a special tool and insert it into the vent of the snake, popping is where you have to squeeze the vent just right to "pop" the snake out if it's a male. Again if you don't know what your doing have someone that does know do this as it's very easy to damage the snake

With probing there is a special tool you by and insert it into the snakes valve, popping is when you squeeze the valve and litterally pop the penis out if it's a male. Neither of these should be done by an inexperienced person as you can seriously harm the snake, a reptile vet is the best person to sex a snake

The best way I know of is to locate the cloaca of the snake. This is like a humans anus. Then take a probe or thin wire and slide it gently into the cloaca, towards the tail. If it goes down into the base of the tail, it is a male snake. If you insert the probe and it stops, its a female.
Male and Female. It's very hard to tell the genders apart as there are no outward signs, there are 2 methods to sexing your snake popping and probing. Popping is done when the snake is very young and probing when the snake is older (NEVER do this if you don't know what your are doing, snakes are easy to damage this way)
