Sorry but it is very difficult to unlock all the characters without using time trials or mirror mode. You can't unlock: Mii outfit B, Funky Kong, Baby Luigi and Toadette. To unlock the others....

Baby Daisy - Attain at least 1 Star Rank for all 50cc Wii Grand Prix Cups
Mii outfit A - earn a trophy in 100cc special cup
Dry Bones - earn a trophy in 100cc leaf cup
Bowser Jr. - Attain at least 1 Star Rank for all 100cc Retro Grand Prix Cups
Daisy - earn a trophy in 150cc special cup
Dry Bowser - Attain at least 1 Star Rank for all 150cc Wii Grand Prix Cups
Birdo - get 250 wins online (usually you would have to play time trials on 16 different courses)
Rosalina - Do something weird with a super Mario galaxy save (usually you would have to attain at least one star rank for all mirror grand prix cups) I am not the person to tell you about how to unlock Rosalina with a Super Mario galaxy save as I don't have super Mario galaxy.

There are many things you can do to unlock things. First off, you can complete Cups in the Grand Prix to earn more tracks, characters and karts. Another thing you can do is to beat staff records in Time Trials, usually the more records you beat, the more characters you unlock. A third way to unlock things is to race. A lot. I mean A LOT!!! If you have thousands apon thousands of races, you can unlock characters without beating any time trials. The fourth and final way to unlock characters is to have certain save data on your Wii. You can unlock 1 secret character if you have save data for Super Mario Galaxy 1 on your Wii's memory. It doesn't even have to be your own save data...you can borrow someone else's memory card.
You have to play as one player, by going to Grand Prix or Time Trials to unlock things such as new characters, vehicles, cups and courses. I hope this answer helps because this is what I do.

The best thing to do to unlock characters is to beat each and every cup, in all difficulties (50, 100, 150 & mirror) in first place.I think there's at least one or two characters that need to be unlocked by beating the normal staff ghosts in the time trials, too.Also, to unlock Mii Suit B, I think you need to either beat every expert staff ghost, and/or get a triple star rating in each cup on all difficulties.
