It depends on the instance. The Nazis made the claim that anyone who had one Jewish grandparent was Jewish. The Israelis have slightly different criteria. The Spanish Inquisition made the claim that someone was a Jew if that person claimed to be a Jew or if they failed to be a "proper" Christian.

A reason for the leading of the Holocaust was based on religion and status of Germany. As for the religious part most people of the Christian fate dwelling in German had come to believe that anyone who was of the Jewish fate had magical powers and were not human. Therefore, the Nazi Party (A government part organization) decide it was best to take out a "threat". Also another drive to the take over kinda had to have been the yearning of having a perfect German land. They took out other people as well homosexuals, elderly, disabled children, ones said to be Jewish, anyone they felt to be unfit.
