That's actually two different things.
Supervisors are nominated and selected by other supervisors.
A Ruby contributor is someone who has made 500,000 contributions, be they a supervisor or not.
No, you have to be a Category Supervisor in order to become a Senior Supervisor. (See the Related question.)
If a floating supervisor wishes to become a category supervisor, they can apply in the same way that a non-supervisor would, by sending an email to the appropriate email address.
A Supervisor can become an Advanced Supervisor and get more tools, but they are not "higher" than regular Supervisors. They can also become a Senior Supervisor, but they don't get additional powers.
You have join one of WikiAnswers groups, and then you will be assigned special tasks, then you will become a supervisor.

Because, being a Supervisor whether it's floating or a category, or a senior Supervisor, it requires a lot of responsibility.

Generally, you don't become a Supervisor based upon on how many contributions you've made to Answers as far as answering questions, editing questions etc., you become a Supervisor based on your personality as far as how you treat others. You could also become a Supervisor a lot easier if you're in the Special Project Assistants program.
Try any supervisor, they can examine your activity and if they believe it is good enough they will recommend you. If the supervisor you contact does not want to do this they can tell other supervisors.
