In the new version of Twitter, click on "@mentions" underneath the text box to see messages that you're mentioned in.
To view your Direct Messages (DM) click on "Messages" at the top of the page, 3 tabs in from the search box.
By clicking on your "Messages" tab from the drop down menu with the face on it. This is located at the top right of the page.

It is impossible to send a DM on Twitter to someone who isn't following you. The user has to follow you before you can send them a DM.

You can DM all people on Twitter. The problem with DM'ing some people is because they have not yet followed you. Until a person follows you back you will remain unable to DM them. Once they have followed you back you can do both public (@) and private (DM) conversations. Unless, of course, they stop following you for whatever reason. The moment they do that you no longer will be able to DM them. Good reason to maintain Twitter etiquette and playing nice on Twitter else you be unfollowed. Or worse, even blocked. If you receive to many blocks your Twitter account can actually be suspended. All part of the way the world works on Twitter.

When a user says they are DM you they are Direct Messageing you. This is a type of private chatting. None of your or their follows can not see the chat
