Pain is caused when an external stimuli disturbs the regular functioning of the physical or internal body. For example, when someone slaps or when a wound is formed after falling down.
The following are some of the ways of managing pains:
1) Medication: Proper medication with drugs such as pain killers are affective as they relieve pain. Eg. Paracetamol.
2) Hypnosis: For those people who can be hypnotized, this method helps them during the process of which the presence of pain will not be felt. It is done by diverting attention to a trance where pain becomes secondary or the pain will not be felt at all.
3) Low voltage currents: This can be another effective way of pain management. When low voltage currents are given to the part where pain exists, it kills the cells which are creating the pain.
4) Surgery: Surgery is the method in which the painful part of the body is cut and thrown off after operating. This is however, the last method to be resorted to, for pain management.
5) Voluntary Attention-Diversion: Another method for managing pain is to divert our thoughts and perception that concentrate of the pain and substitute one's attention with other pleasant thoughts and perception.
There are other methods as well in physiotherapy and exercise techniques for managing pain.

Other meds such as anti-depressants can sometimes be used to minimize the perception of pain, but generally do not work to "kill" pain when it is truly being felt, as with an injury or chronic pain condition.
The exact method of pain relief depends upon which treatment is administered. For example, if you take pain pills, the chemicals in the meds act on the receptor sites to either block the body's interpretation of pain or induce a sense of well-being, otherwise known as euphoria. That is what can make that treatment dangerous to addictive personalities if not monitored carefully.
NSAIDS or steriods may reduce inflammation, enabling the healing or tissue, hence, pain will lessen as the nerves are no longer under intense pressure.
Of course, this is all a very simple explanation and a pharmacist or physician could better explain it more scientifically.
