You can't send a private message to someone who isn't following you, but you can send a reply using a at (@) followed by the person's username and your message. You can do this by Mentioning them or by replying to one of their tweets.
Start your tweet with @username (replacing 'username' with their actual username) Your tweet will then appear in their 'mentions' tab.
You send them an @reply by typing the @ symbol then, the users name, then the message you wish to send.
Type their username into your tweet preceded with an "@"
Simply begin your tweet with their username (@username) and your tweet will appear in their @replies tab.
By Mentioning that person username as @username or by replying to one of his/her tweets.

If your account is private then only people that are following you can see your tweets. It doesn't matter who you are following, they must be following you to view your tweets. So, if you are following someone but they aren't following you then they won't be able to view your tweets.
The only way you would receive these tweets is if they are retweets or if you are following them.
By following many people, most of whom will follow you in return. You can then send tweets to them personally, or to everyone.
If your tweets are protected then only people following you can see your tweets. If she isn't following you then maybe she has access to an account that is following you.
If you protect your updates, the people who follow you CAN see your tweets, but ONLY the people who follow you.
You may send a tweet (or @reply) to anyone on Twitter; you certainly don't have to be following them! Following a Twitter user just means that all their tweets appear in the "timeline" on the home page of Twitter when you log in. (Along with your tweets and the tweets of anyone else you may be following.) To follow someone, go to their Twitter page and click the "follow" button. Anyway, to send a tweet to any Twitter user, just include an @ sign followed by the username of the user you want to send the tweet to. For example, if you want to send a tweet to the user with the username JohnSmith, include @JohnSmith in you tweet.
The only way that your tweets will appear on their timeline is if they are following you. If you want someone to see a tweet but they aren't following you then you should 'mention' them in your tweet (@username)
