There is a camera glitch you can use to get out; and if you have a companion with you firing them will cause them to open the door themselves.

You can hear it after you beat the quest "Project Purity." You will hear a distress signal from the vault and if you listen to it you will get a new quest, "trouble on the homefront" once you walk into the door of the vault you will once again hear the Vault 101 PA system, not that there's much to hear. p.s. you have to be somewhere near the vault such as in Megaton to hear the distress beacon.

You can not go back in and out of Vault 101. After you first leave the Vault the only time you can go back in is for the quest "Trouble On The Homefront." but once you beat that side quest you are not allowed back in Vault 101 ever.
