There is no way of getting a female wolf pregnant unless you play through the single player game. You won't see any signs of a she-wolf being pregnant; she will perform as she would normally and you won't see any scenes relating to her being pregnant/giving birth regardless of her being your wolf or your mate. Naturally this was deliberately left out to maintain a family-friendly game that could be played by all age groups.
That being said, you won't see a birth scene or signs of her carrying pups. They are born after completing the 'Mark your Territory' mission.
If you're referring to a she-wolf being pregnant in multiplayer, it's not possible due to pups being locked to the single player campaign. The focus of WolfQuest is to be educational. As such, mating/dating is not allowed in multiplayer.
On the 1st WolfQuest you can't... However on WolfQuest 2 you can but you need a mate on the 1st one.

There is no current way to enter your den in WolfQuest. Wolf dens are normally very small and only pregnant wolves (and sometimes a few trusted other females) will only enter it.
