Keep it short. A few minutes at the most. Nobody wants to hear a life story. The question really means they want to know about you and how your experience relates to the job. They aren't interested in where you grew up, your kids, dogs and cats, or your weekend hobbies. I say that because I've actually heard of people answering that way.
For example, let's say you are interviewing for a customer service job handling billing escalations. There is not enough money in the Federal Reserve for me to take a job like that, but this isn't about me. So anyway, you go to the interview, and you get asked "Tell me about yourself?" Your reply should be 1-3 minutes, and you tell about the jobs and any experience you had in the past, even volunteer work. How long have you done this kind of work, what size companies have you worked for, promotions, special characteristics (maybe you resolved more complaints than anybody else on your team), any recommendations or recognition you received, and what you are bringing to the company, and how they will benefit. Remember, you have to make it about them to start with. All they are interested in is what YOU can give them. Businesses are very selfish that way.
Can that be done in just a few minutes? Yes it can. As with anything on your resume, when they ask any question, you aren't going into the full history of it, you are giving a brief overview, and if they want to know more, they will ask. Which is why you limit answers to just a few minutes at the most.
So that is a self introduction. You are telling them briefly why you are there, your skills, experience, what you have done in the past that relates, and leaving yourself open for more questions.
Practice with family or friends. See how long it takes to answer. Remember the purpose of an interview is to sell yourself.

Call the employer the day before the interview and tell him that the interview needs to cancelled because of conflict in schedule. Discuss how the interview can be rescheduled. In case of acceptance to another job, call the employer again and inform him of the new job taken.
